I know we're working on 11 right now, but I'm going to put up a description of what Georgia, L and I have for Lily so far.

She meets the 9th Inspector in the opening episode of the new!IS series. She's a very clever young woman. At first the Inspector doesn't respect her so much as he finds that she's useful/he can put up with her easily. So he takes her with him to be an associate. He eventually grows to care for her in his own way and she has an obvious crush on him (which doesn't interfere with her decision making so the Inspector finds he doesn't mind her feelings). Some time before 9 dies, she kisses him. He doesn't reciprocate but he also doesn't push her away.
His next regeneration and her become rather good friends. He'll often share inside jokes with her and ridicule people to make her giggle. He trusts her but unfortunately at some point in the series she's separated from him for good (somehow, we don't have that planned yet). Before he loses her he does kiss her, though it's not one of those sexually charged TV kisses. It's more meant as a "Look this is an emotional gesture right? This is how I show I care, right?" sort of move on the Inspector's part (the impact of which is achingly touching for the audience).
After she leaves 10 is more moody than before and begins travelling a lot more frequently with the Constable. There's eventually a big Inspector vs Villain fight and at the end of it the Inspector is ready to die. He never says it out loud but the audience knows that part of the reason is because Lily is gone. His bond with her helped make 10 what he is; she's a part of him, as much as he can accept someone else emotionally, and without her he's ready to move on. It's not that he's still torn up about it or depressed or jaded with his life now, it's simply that without Lily there's no where left for that incarnation to go; there's no growth to be had. So when the times comes, he finds he's ready and accepts his death with grace (and just a hint of melancholy, because deep down he does miss her).
